Survival strategies of unicellular phototrophs: using microalgae to study photosynthesis, energy storage and stress adaptation

Microalgae are found in diverse environments from soil, marine waters, to alpine snowfields. Ecologically, they are an important group of organisms as they are one of the main primary producers in aquatic environments and are responsible for a vast amount of oxygen produced on this planet. Being unicellular, microalgae are constantly exposed to changing environments and must rapidly adjust its cell physiology accordingly and establish long-term survival strategies when conditions become suboptimal. Photosynthesis is the main metabolic driving force in these phototrophs and must constantly be monitored and adjusted. In order to survive stressful conditions (such as low nutrients), microalgae can photosynthetically assimilate carbon in the form of oil (mostly triacylglycerols) as intracellular high-energy reserves. Regulatory mechanisms for photosynthesis and oil accumulation are not yet fully understood. Increasing our understanding of these cellular processes would be valuable for understanding the effects of environmental changes in microalgal metabolism, but also for biotechnological applications, as algae are used for industrial oil production. Chlamydomonas reinhardtii is a unicellular freshwater green alga that has been used to study many basic components of photosynthesis. C. reinhardtii is a good model system to study these bioenergy-relevant mechanisms because there are many molecular tools already established.
In this talk, I will first present work on understanding the regulation of photosynthesis under anaerobic stress conditions, where we found new components of photosynthetic complexes. Secondly, I will present a high-throughput screen method that we have developed for isolating mutants with increased oil accumulation in C. reinhardtii. Finally, I will discuss plans for future research where I will investigate the survival mechanisms of snow microalgae found in Antarctica and alpine snowfields and its interaction with this unique psychrophilic microbial community.
