南極大陸ラングホブデ:極域生態系における微生物進化適応

今朝、久しぶりにJAMSTECの高野博士にお会いいたしました。低温科学研究所で開催されている研究集会に参加されているとのこと。本当に懐かしい限りです。

つい先日、南極大陸ラングホブデから単離した微生物のゲノムに関する論文が受理されたばかりでしたので、彼との再会は一層嬉しく感じられました。

2006年1月末、昭和基地からおよそ37km離れた露岩地帯ラングホブデで研究観測を行いました。メンバーは総勢3名で、高野さんと環境省の桝厚生さん。約2週間、彼らと楽しく、かつ充実した日々。



ラングホブデは、南極特別保護地区に指定され、立ち入りが制限されています。許可を得て研究観測を進めていたところ、思わぬ発見が多々ありました。



その一つが、赤雪現象です。帰国後、高野さんの窒素安定同位体比の解析結果は驚くものでありました。微生物群衆構造解析、赤雪藻類の色素分析等の結果を含めて、発表した論文は下記の通り。

今回、ラングホブデの観測小屋近くで採取した海洋沿岸堆積物から発見した新規微生物ですが、まだ未記載です。ゲノム解析が進み、先に発表することになりました。きっと、この菌のゲノム情報は、南極海洋生態系において微生物の進化・環境適応を解明する上で重要になると思われます。

東京湾の青潮

このところ、東京湾で青潮発生。その発生メカニズムは、いささか複雑なのですが、大雑把に言うと下記の通り。

生活排水等が東京湾へ流入(栄養塩等の流入)

春から夏にかけて海水温上昇

海水表層で植物プランクトンの増殖

プランクトンの枯死による海水深層へ沈降

深水層や底泥で有機物の好気的微生物分解

深水層の貧酸素化

底泥や深水層での微生物学硫酸還元による硫化水素発生

強風発生により深水層中の硫化水素が表層水へ巻き上がり

酸素による硫化水素自己酸化で元素状硫黄の生成
(結果として青潮の発生)

地下生物圏で活躍する主役たち:SulfuritaleaとSulfuricella

地球上の様々な環境に微生物が生息していて生態系を構築していますが、その微生物を人為的に培養しようとしてもほとんどが困難です。これを「微生物のダークマター」と呼んでいますが、この問題を克服するためのアプローチとしてメタオミクス手法(メタゲノム、メタプロテオーム、メタトランスクリプトームなど)が近年急速に発達しています。

さて、南アフリカのウィットウォーターズランド盆地地帯は世界最大級の金鉱床としても知られています。その地下1.3kmという深い層では、スライム(SLiMEs; subsurface lithoautotrophic microbial ecosystems)と呼ばれる無機独立栄養微生物生態系が形成されていることが知られています。温度38℃、pH8.6、汽水性かつ貧酸素環境です:水素濃度8.9nM、メタン濃度1.3mM、硫酸塩濃度623μM、硫化物濃度14.9μM。

プリンストン大学の研究グループが調べたところ、このスライムは脱窒硫黄酸化によって駆動されており、その主役がSulfuritalea(サルフリタレア)とSulfuricella(サルフリセラ)であることが明らかにされました。これらの菌は、小島久弥さんが山梨県のみずがき湖から単離し、新種として記載したものです。

こうしたメタオミクスを駆使した研究が進展するほど、生態系を構成している微生物の単離培養の重要性がいっそう増すことが再認識されます。

それにしても、Sulfuritalea(サルフリタレア)とSulfuricella(サルフリセラ)。私たちの予想を超えて、さまざまな生態系での報告が増えて来ています。その下支えになっていることの一つは、両菌の完全長ゲノム解読にも成功していることです。

改めて、小島さんの不断の努力に脱帽です。

残雪期の山岳調査

今シーズン最初の山岳調査。天候条件は良いとは言えませんが、総勢4人で現場へ。

残雪期の山岳地帯での調査は危険を伴うので、低温科学研究所技術部の森さんにお願いして、安全第一で調査を行なっています。今回、大学院生のAさんは初めての経験と言うことで、行動開始前に森さんより野外行動技術講習です。



さあ、調査地点へ。





助教の寺島美亜さんは、Aさんに試料採取や現場観測法など丁寧に教えています。Aさんも初めてではありますが、熱心かつ意欲的に取り組んでいます。

Aさんのご好意で、全自動型カフェマシーンがオフィスに設置されました。オフィスのオシャレ度が格段にアップした感があります。



先週の金曜日と土曜日、それぞれ、寺島さんがイギリス・アバディーンから、私がスイスのバーゼルから帰国。お土産と共に味わい深いカフェを頂く。リッチな気分転換で仕事も捗りそうです。

小宇宙としての湖:淡水の貧酸素•無酸素水塊での微生物たち

2004年8月に低温科学研究所に赴任してから13年が過ぎようとしています。新天地での研究展開をいかに行うか? 2005年4月に赴任した小島久弥さんと議論し、研究フィールドの一つとして山梨県のダム湖である「みずがき湖」を選びました。

山梨大学の岩田智也先生のご協力のもと、2006年より現場での調査を開始。みずがき湖は、標高800mにあり、最大水深52mで、成層構造が発達します。水深25m以深は、常に低温かつ貧酸素ないし無酸素状態が維持されています。このような環境で物質循環はどのように行われるのか、それを担う微生物の生理生態は? 

幸運にも期待以上の成果が得られ、驚きの連続です! 一連の研究には、小島久弥さんの貢献もさることながら、現在ドイツでアレキサンダーフンボルトフェローとして活躍中の渡邉友浩さんの存在も大きかったと思います。 

今後もワクワクする新発見が続出しそうで、楽しみです。
