3月16日のＮＨＫ Ｅテレ「緊急SP！?STAP細胞の謎に迫れ」。
冒頭に「今この時期に本当にやるの？」という声がありそうですが、
やります、サイエンスＺＥＲＯですから、
という話が。
ですが、小保方晴子さんの博士論文が切り貼りかどうか、という話と、
ＳＴＡＰ細胞が本当に存在するのか、という話を
切り分けて考えたい、とは私も思いますので、
変更や中止にならなくてよかったです。
備忘録を兼ねて、番組の内容をメモしておきます。
まず、
理研の野依良治理事長が謝罪する映像が流れ、
英国Nature誌に論文が発表されて世界中の注目を集めた。
しかし、

世界中の専門家が再現を試みても成功しない
→
論文に証拠として示された画像に疑いがもたれる
→
日本分子生物学会理事長が緊急声明を出した。
→
論文の撤回が迫られるに至った
という風に経緯を説明。
ＳＴＡＰとは？
ＳＴＡＰ細胞

細胞に酸などの刺激を与えると、
初期化されて、あらゆる組織の細胞になる可能性を獲得する、
本来、細胞はあらゆる種類の細胞になることができるが、
ある組織の細胞に分化しはじめると、
他の細胞になる遺伝子にはフタがされた状態になる。
初期化とは、そのフタを外して、
再び、他の種類の細胞になる可能性を獲得させる、というもの。
Ipｓ細胞は、一度分化した細胞にいくつかの遺伝子を入れる、という
難しい方法をとるが、
ＳＴＡＰ細胞は、酸で刺激する、という簡単な方法であることが
注目された。
しかし、14日の理研の記者会見では、
論文撤回に言及される事態に至っている。
しかし、「サイエンスＺＥＲＯ」はまだ中間報告なので、どうなるのかは
分からない、と慎重な姿勢。
ＳＴＡＰ細胞のポイントは、
ＯＣＴ４と、ＴＣＲ再構成だ、としてこの２つについて解説。
【ＯＣＴ４とは？】
ＯＣＴ４は、可能性細胞の目印、といえる細胞。
研究チームは
酸に漬けた細胞にＯＣＴ４が現れると緑色にヒカル細胞を持ったマウスを用意。
そのマウスの細胞を、酸に漬けて処理し、
培養して2日後から、緑色の細胞が現れ始めた。
この細胞は、本当にあらゆる細胞に分化できるのか、を確認するために、
細胞をマウスの皮膚に注射した。
すると腫瘍ができ、
腫瘍から、皮膚、筋肉、腸などの細胞が発見された。
しかし、この段階では、また
ips細胞のような万能細胞ができたわけではない。
ＳＴＡＰ細胞自身は自分では増殖できず、
死んでしまう。
マウスに移植して生体の中で増えるだけでは万能細胞とは言えない。
そこで研究チームは、
あるホルモンを加えて、増殖能力を得た細胞を
ＳＴＡＰ幹細胞とした。
しかし、国内外の研究者が追試をしても、
再現性がない、と言われるようになった。
発表直後から追試を行っている
関西学院大学 関由行さんを取材した。
細胞の中の初期化についての研究者である関さん。
論文には、血液細胞だけではなく、
いろいろな組織の細胞からできた、と書かれていたので、
「けっこう、あっさりできるのかな」と考え、
ＰＨ５．７の酸性の液体につけて実験を行ったが、
３回の実験で、すべて細胞は死んでしまった。
番組に出演した、
八代 嘉美 京都大学 iPS細胞研究所特定準教授は
発表された手順通りにやっても、
人の手が違ったり、実験器具のメーカーが違うと失敗することもある、と説明。
そこで、研究者達から、
詳細な実験手順を公開して欲しい、という声が上がり、
理研は詳細な手順書を公開した。
ところが、
理化学研究所が発表した詳細な手順書の中の一文が
さらに物議を醸すことになった。
その一文とは、
「ＳＴＡＰ幹細胞にはＴＣＲ再構成がなかった」
というもの。
【ＴＣＲ再構成の有無】
ＴはＴ細胞（病原体などから体を守る働きを持つ）のことで
様々なレセプターを持つ。
多様な病原体に対応するため、
ＤＮＡの一部を切りとって再構成される、
という他の細胞にない可能性を持つ。
ＴＣＲ再構成＝Ｔ細胞に分化した証
ＳＴＡＰ細胞は、酸に漬けたことによって
一度分化した細胞が初期化されて、
その後、再度どんな細胞になることもできる可能性を持つ、
という事なのですが、
酸に漬けた細胞の中にはＴＣＲ再構成が含まれていたので、
ＳＴＡＰ細胞に
ＴＣＲ再構成が認められれば、
その細胞は、
もともとＴ細胞だったものが、初期化されたものである、
ということになる。
理研が公開した実験の手順書には
ＴＣＲ再構成はなかった、と書かれていたが、
ＳＴＡＰ細胞ではあったが、
ＳＴＡＰ幹細胞に変わるときに、
ＴＣＲ再構成の遺伝子を持っている細胞は生存が不利になるので、
無くなってしまう、という説明だった。
さらに14日の記者会見では、
ＳＴＡＰ細胞にも、本当にＴＣＲ再構成があったのか？
について、
その根拠として論文に掲載された写真が、
別の実験からの切り貼りであることを
理化学研究所が認めたため、
ＳＴＡＰ細胞でＴＣＲ再構成ができた、ということに対する信用性が揺らいでいる。
可能性１
本当にＳＴＡＰ細胞にはＴＣＲ再構成ができて、
ＳＴＡＰ幹細胞になるときに消えてしまった
可能性２
酸で処理することによって、
もともと存在した可能性細胞が選択されている
マウスの皮膚にＳＴＡＰ細胞を注射したら、
腫瘍ができた、
その腫瘍の中には、
皮膚、筋肉、腸、という異なる細胞が存在する、
としていた証拠の写真が、
小保方晴子さんが、
学位論文に掲載した
骨髄から取り出した細胞をマウスに注射して得られた画像と
同じ物だった。
共同執筆者の若山教授が、
小保方さんから渡されたＳＴＡＰ幹細胞を切りとって、
培養したところ、
１匹のマウスになった。
しかし、渡された細胞がなんだったのか、
若山さんには分からない。
このような論文が発表されてしまった原因は？
八代準教授は、
さまざまな分野の専門家が集まって研究しているので、
他のジャンルの専門家が持ってきたデータを、
あからさまに疑うことは難しい、と指摘した。
では、人間の細胞からはＳＴＡＰ細胞のようなものはできないのか？
シクラメンの茎に「切る」という刺激を加えると、
切り口に、
カルスという初期化した細胞の固まりができる。
その細胞を培養すると、
根、葉、花、
などのすべての組織ができる。
植物でできることが、人間の細胞で絶対に起きない、
とはいえないので、ＳＴＡＰ細胞のような細胞が存在する可能性はないわけではない。
しかし、今回のＳＴＡＰ細胞の論文については、
さまざまな問題があり、
日本の科学に対する信頼が揺らいでしまったのではないか、
はっきりとした決着をつけて欲しい、というコメントで結ばれました。
