There is a significant medical need for tough biodegradable polymer adhesives that can adapt to or recover from various mechanical deformations while remaining strongly attached to the underlying tissue. We approached this problem by using a polymer poly(glycerol-co-sebacate acrylate) and modifying the surface to mimic the nanotopography of gecko feet, which allows attachment to vertical surfaces. Translation of existing gecko-inspired adhesives for medical applications is complex, as multiple parameters must be optimized, including: biocompatibility, biodegradation, strong adhesive tissue bonding, as well as compliance and conformability to tissue surfaces. Ideally these adhesives would also have the ability to deliver drugs or growth factors to promote healing. As a first demonstration, we have created a gecko-inspired tissue adhesive from a biocompatible and biodegradable elastomer combined with a thin tissue-reactive biocompatible surface coating. Tissue adhesion was optimized by varying dimensions of the nanoscale pillars, including the ratio of tip diameter to pitch and the ratio of tip diameter to base diameter. Coating these nanomolded pillars of biodegradable elastomers with a thin layer of oxidized dextran significantly increased the interfacial adhesion strength on porcine intestine tissue in vitro and in the rat abdominal subfascial in vivo environment. The Lower Cretaceous Tetori Group of Japan has yielded diverse freshwater and terrestrial vertebrate assemblages. The most productive small vertebrate locality is the ‘Kaseki-Kabe’ or ‘fossil-bluff’ at Kuwajima, Hakusan City, Ishikawa Prefecture. These deposits have produced at least six distinct lizard taxa of which one, described and named here as Kuwajimalla kagaensis, has lanceolate denticulate teeth convergent on those of the living Iguana. This type of dentition is rare among living lizards and is usually considered indicative of herbivory and, more specifically, folivory. Kuwajimalla kagaensis provides the earliest unambiguous record of squamate herbivory. Comparisons with modern and fossil lizards suggest that Kuwajimalla may be an early relative of the macrocephalosaurines, a group of large herbivores well represented in the Upper Cretaceous of Mongolia.# asahi.comが雑誌名をきちんと示すなんて珍しい
Fertile queens and sterile workers are alternative forms of the adult female honey bee that develop from genetically identical larvae following differential feeding with royal jelly. We now show that silencing the expression of DNA methyltransferase Dnmt3, a key driver of epigenetic global reprogramming, in newly hatched larvae, leads to a stunning royal jelly-like effect on the larval developmental trajectory; the majority of Dnmt3 siRNA-treated individuals now emerge as queens with fully developed ovaries. Our results suggest that; DNA methylation in Apis is utilized for storing epigenetic information; that the utilization of that information can be differentially altered by nutritional input, and that the flexibility of epigenetic modifications underpin profound shifts in developmental fates with massive implications for reproductive and behavioural status.# それって、ローヤルゼリーを食べて続けるとDNAのメチル化状態が変わるってこと?
Intestinal adhesions are bands of fibrous tissue that connect the loops of the intestine to each other, to other abdominal organs or to the abdominal wall. Fibrous tissue formation is regulated by the balance between plasminogen activator inhibitor type 1 (PAI-1) and tissue-type plasminogen activator (tPA), which reciprocally regulate fibrin deposition. Several components of the inflammatory system, including cytokines, chemokines, cell adhesion molecules and neuropeptide substance P, have been reported to participate in adhesion formation. We have used cecal cauterization to develop a unique experimental mouse model of intestinal adhesion. Mice developed severe intestinal adhesion after this treatment. Adhesion development depended upon the interferon-γ (IFN-γ) and signal transducer and activator of transcription-1 (STAT1) system. Natural killer T (NKT) cell–deficient mice developed adhesion poorly, whereas they developed severe adhesion after reconstitution with NKT cells from wild-type mice, suggesting that NKT cell IFN-γ production is indispensable for adhesion formation. This response does not depend on STAT4, STAT6, interleukin-12 (IL-12), IL-18, tumor necrosis factor-α, Toll-like receptor 4 or myeloid differentiation factor-88–mediated signals. Wild-type mice increased the ratio of PAI-1 to tPA after cecal cauterization, whereas Ifng-/- or Stat1-/- mice did not, suggesting that IFN-γ has a crucial role in the differential regulation of PAI-1 and tPA. Additionally, hepatocyte growth factor, a potent mitogenic factor for hepatocytes, strongly inhibited intestinal adhesion by diminishing IFN-γ production, providing a potential new way to prevent postoperative adhesions.# 開腹手術時の癒着が防げたら色んな面で助かることが多いですね
