財布を拾う

昨夜、YK外勤から帰宅しまして、車を停めて玄関に入ろうとすると…。

家の真ん前の信号付き横断歩道前に何かが落ちています。はて?ここにこんな出っ張りあったっけ?と、近寄ってみると明らかに財布…。雨で表は多少濡れてはいますが、裏は乾いていて、落としてから間もない様子…。仕方なく、拾い上げて玄関に入りまして、家の中で財布の中に何か連絡先がないか調べました。が、あるのは現金と銀行のキャッシュカード、運転免許証で、電話番号はなし。落とし主は若い男性です。困っているだろうなぁ…。まぁ、直接連絡出来ないけど、警察に届ければ持ち主には戻るだろうということで、再び車を出して最寄りの交番に届けて来ました。

最近は、落とし主に拾い主の連絡先を教えてよいかを予め決めておくそうで、落とし主に気を使ってもらいたくないので、開示不可にしてきました。あとから、思うと、無事に落とし主の手元に戻ったかどうかが心配になったので、それだけでも教えてもらえるように頼めばよかったと、後悔しましたが、まぁ、大丈夫でしょう。

教訓:財布など大事なものは連絡先が分かるようにしておきましょう。

昨日、仕事場にやってきたTH&MH夫妻宅のお嬢様。骨格はお父さん似だけどパーツはお母さん似?

会社内の別センターで実験材料になってきた(え?書き方悪いって?なんて書けばいいんだ?)そうです。0歳児にも関わらず、既にお金を稼いでいます。将来が楽しみというか末恐ろしいというか…

あ、ちなみにお腹が空いて大泣きした挙げ句にお腹がふくれたあとに爆睡してました。ま、それが本来のお仕事ですからね。

    Somatic cells can be reprogrammed to a pluripotent state through the ectopic expression of defined transcription factors. Understanding the mechanism and kinetics of this transformation may shed light on the nature of developmental potency and suggest strategies with improved efficiency or safety. Here we report an integrative genomic analysis of reprogramming of mouse fibroblasts and B lymphocytes. Lineage-committed cells show a complex response to the ectopic expression involving induction of genes downstream of individual reprogramming factors. Fully reprogrammed cells show gene expression and epigenetic states that are highly similar to embryonic stem cells. In contrast, stable partially reprogrammed cell lines show reactivation of a distinctive subset of stem-cell-related genes, incomplete repression of lineage-specifying transcription factors, and DNA hypermethylation at pluripotency-related loci. These observations suggest that some cells may become trapped in partially reprogrammed states owing to incomplete repression of transcription factors, and that DNA de-methylation is an inefficient step in the transition to pluripotency. We demonstrate that RNA inhibition of transcription factors can facilitate reprogramming, and that treatment with DNA methyltransferase inhibitors can improve the overall efficiency of the reprogramming process.

    The extinct placoderm fishes were the dominant group of vertebrates throughout the Middle Palaeozoic era, yet controversy about their relationships within the gnathostomes (jawed vertebrates) is partly due to different interpretations of their reproductive biology. Here we document the oldest record of a live-bearing vertebrate in a new ptyctodontid placoderm, Materpiscis attenboroughi gen. et sp. nov., from the Late Devonian Gogo Formation of Australia (approximately 380 million years ago). The new specimen, remarkably preserved in three dimensions, contains a single, intra-uterine embryo connected by a permineralized umbilical cord. An amorphous crystalline mass near the umbilical cord possibly represents the recrystallized yolk sac. Another ptyctodont from the Gogo Formation, Austroptyctodus gardineri, also shows three small embryos inside it in the same position. Ptyctodontids have already provided the oldest definite evidence for vertebrate copulation, and the new specimens confirm that some placoderms had a remarkably advanced reproductive biology, comparable to that of some modern sharks and rays. The new discovery points to internal fertilization and viviparity in vertebrates as originating earliest within placoderms.

    The HIV-1 Vpu protein is required for efficient viral release from human cells. For HIV-2, the envelope (Env) protein replaces the role of Vpu. Both Vpu and HIV-2 Env enhance virus release by counteracting an innate host-cell block within human cells that is absent in African green monkey (AGM) cells. Here we identify calcium-modulating cyclophilin ligand (CAML) as a Vpu-interacting host factor that restricts HIV-1 release. Expression of human CAML (encoded by CAMLG) in AGM cells conferred a strong restriction of virus release that was reversed by Vpu and HIV-2 Env, suggesting that CAML is the mechanistic link between these two viral regulators. Depletion of CAML in human cells eliminated the need for Vpu in enhancing HIV-1 and murine leukemia virus release. These results point to CAML as a Vpu-sensitive host restriction factor that inhibits HIV release from human cells. The ability of CAML to inhibit virus release should illuminate new therapeutic strategies against HIV.

    Musashi1 (Msi1) is an RNA-binding protein that is highly expressed in neural stem cells. We previously reported that Msi1 contributes to the maintenance of the immature state and self-renewal activity of neural stem cells through translational repression of m-Numb. However, its translation repression mechanism has remained unclear. Here, we identify poly(A) binding protein (PABP) as an Msi1-binding protein, and find Msi1 competes with eIF4G for PABP binding. This competition inhibits translation initiation of Msi1's target mRNA. Indeed, deletion of the PABP-interacting domain in Msi1 abolishes its function. We demonstrate that Msi1 inhibits the assembly of the 80S, but not the 48S, ribosome complex. Consistent with these conclusions, Msi1 colocalizes with PABP and is recruited into stress granules, which contain the stalled preinitiation complex. However, Msi1 with mutations in two RNA recognition motifs fails to accumulate into stress granules. These results provide insight into the mechanism by which sequence-specific translational repression occurs in stem cells through the control of translation initiation.

    One-to-one orthologous genes of relatively closely related species are widely assumed to have similar functions and cause similar phenotypes when deleted from the genome. Although this assumption is the foundation of comparative genomics and the basis for the use of model organisms to study human biology and disease, its validity is known only from anecdotes rather than from systematic examination. Comparing documented phenotypes of null mutations in humans and mice, we find that >20% of human essential genes have nonessential mouse orthologs. These changes of gene essentiality appear to be associated with adaptive evolution at the protein-sequence, but not gene-expression, level. Proteins localized to the vacuole, a cellular compartment for waste management, are highly enriched among essentiality-changing genes. It is probable that the evolution of the prolonged life history in humans required enhanced waste management for proper cellular function until the time of reproduction, which rendered these vacuole proteins essential and generated selective pressures for their improvement. If our gene sample represents the entire genome, our results would mean frequent changes of phenotypic effects of one-to-one orthologous genes even between relatively closely related species, a possibility that should be considered in comparative genomic studies and in making cross-species inferences of gene function and phenotypic effect.

    Observations of individual virions in live cells have led to the characterization of their attachment, entry and intracellular transport. However, the assembly of individual virions has never been observed in real time. Insights into this process have come primarily from biochemical analyses of populations of virions or from microscopic studies of fixed infected cells. Thus, some assembly properties, such as kinetics and location, are either unknown or controversial. Here we describe quantitatively the genesis of individual virions in real time, from initiation of assembly to budding and release. We studied fluorescently tagged derivatives of Gag, the major structural component of HIV-1―which is sufficient to drive the assembly of virus-like particles―with the use of fluorescence resonance energy transfer, fluorescence recovery after photobleaching and total-internal-reflection fluorescent microscopy in living cells. Virions appeared individually at the plasma membrane, their assembly rate accelerated as Gag protein accumulated in cells, and typically 5–6 min was required to complete the assembly of a single virion. These approaches allow a previously unobserved view of the genesis of individual virions and the determination of parameters of viral assembly that are inaccessible with conventional techniques.

    Mosquito repellency data on acylpiperidines derived from the U.S. Department of Agriculture archives were modeled by using molecular descriptors calculated by CODESSA PRO software. An artificial neural network model was developed for the correlation of these archival results and used to predict the repellent activity of novel compounds of similar structures. A series of 34 promising N-acylpiperidine mosquito repellent candidates (4a–4q') were synthesized by reactions of acylbenzotriazoles 2a–2p with piperidines 3a–3f. Compounds (4a–4q') were screened as topically applied mosquito repellents by measuring the duration of repellency after application to cloth patches worn on the arms of human volunteers. Some compounds that were evaluated repelled mosquitoes as much as three times longer than N,N-diethyl-m-toluamide (DEET), the most widely used repellent throughout the world. The newly measured durations of repellency were used to obtain a superior correlation equation relating mosquito repellency to molecular structure.

    We experimentally demonstrate a simple scheme for generating a four-photon entangled cluster state with fidelity over 0.860±0.015. We show that the fidelity is high enough to guarantee that the produced state is distinguished from Greenberger-Horne-Zeilinger, W, and Dicke types of genuine four-qubit entanglement. We also demonstrate basic operations of one-way quantum computing using the produced state and show that the output state fidelities surpass classical bounds, which indicates that the entanglement in the produced state essentially contributes to the quantum operation.

    Background
    The y-larva, a crustacean larval type first identified more than 100 years ago, has been found in marine plankton samples collected in the arctic, temperate and tropical regions of all oceans. The great species diversity found among y-larvae (we have identified more than 40 species at our study site alone) indicates that the adult organism may play a significant ecological role. However, despite intense efforts, the adult y-organism has never been identified, and nothing is therefore known about its biology.

    Results
    We have successfully and repeatedly induced metamorphosis of y-larvae into a novel, highly reduced juvenile stage by applying the crustacean molting hormone 20-HE. The new stage is slug-like, unsegmented and lacks both limbs and almost all other traits normally characterizing arthropods, but it is capable of vigorous peristaltic motions.

    Conclusions
    From our observations on live and preserved material we conclude that adult Facetotecta are endoparasitic in still to be identified marine hosts and with a juvenile stage that represents a remarkable convergence to that seen in parasitic barnacles (Crustacea Cirripedia Rhizocephala). From the distribution and abundance of facetotectan y-larvae in the world's oceans we furthermore suggest that these parasites are widespread and could play an important role in the marine environment.

    Neural stem cells (NSCs) have attracted considerable attention as a potential source of cells for therapeutic treatment of impaired areas of the central nervous system. However, efficient and clinically feasible strategies for expansion of the endogenous NSC pool are currently unavailable. In this study, we demonstrate that mood stabilizing drugs, which are used to treat patients with bipolar disorder, enhance the self-renewal capability of mouse NSCs in vitro and that this enhancement is achieved at therapeutically relevant concentrations in the cerebrospinal fluid. The pharmacological effects are mediated by the activation of Notch signaling in the NSC. Treatment with mood stabilizers increased an active form of Notch receptor and upregulated its target genes in neural stem/progenitor cells, whereas co-culture with γ-secretase inhibitor or the presence of mutation in the presenilin1 gene blocked the effects of mood stabilizers. In addition, chronic administration of mood stabilizers expanded the NSC pool in the adult brain, which subsequently increased the cell supply to the olfactory bulb. We suggest that treatment with mood stabilizing drugs could be used to facilitate regeneration following insult to the central nervous system.

    Mail.appがぐだぐだ…

    先週の終わり頃から、Mail.appが絶不調…

    何が絶不調かって、メールサーバとの接続だの、メールのダウンロードだの、メールの判別だの、何だのかんだのとやたら時間がかかる上に、一旦それを始めると、一切メールチェックすらしなくなる…。で、終了しようにも通常操作では終了出来ず、仕方なく強制終了…。そんなことの繰り返し。おかげで、急ぎのメールが来ていたことを数時間もあとに知ったり、来るはずのメールが来ないので諦めていたら実はとうの昔に届いていたりと、結構エライ目に遭っている…

    仕方なく復旧作業に入るも、何をやっても一向に回復せず。ライブラリ内のMailフォルダからメールアカウントのローカルデータを外して作り直させたり、Envelope Indexを作り直させたり(これはいちいち時間がかかる)、MessageRules.plistを捨ててルールを一から作成し直したり…。なのに未だに復旧の目処立たず…。どうもミョーなのが、ルールをリセットしたにも関わらず、昔のルールに従ってメールがフォルダに振り分けられた挙げ句に元の受信ボックスにも同じものが残っていたりすること。つまりメールが重複してしまう…。こればかりは非常に具合が悪い…。片付けたと思った仕事が再び現れるようなもので、かなり効率が悪い…。いったいどこに前の設定が残っているのやら…

    どうしたらいいんだ?誰か助けてちょ…

    Context. Gamma-ray burst (GRB) emission is believed to originate in highly relativistic fireballs.
    Aims. Currently, only lower limits were securely set to the initial fireball Lorentz factor Γ0. We aim to provide a direct measure of Γ0.
    Methods. The early-time afterglow light curve carries information about Γ0, which determines the time of the afterglow peak. We have obtained early observations of the near-infrared afterglows of GRB 060418 and GRB 060607A with the REM robotic telescope.
    Results. For both events, the afterglow peak could be clearly singled out, allowing a firm determination of the fireball Lorentz of Γ0 ∼ 400, fully confirming the highly relativistic nature of GRB fireballs. The deceleration radius was inferred to be Rdec ≈ 1017 cm. This is much larger than the internal shocks radius (believed to power the prompt emission), thus providing further evidence for a different origin of the prompt and afterglow stages of the GRB.

    Objective: To investigate the association of dietary folate, vitamin B6 (VB6) and vitamin B12 (VB12) with the risk of coronary heart disease among middle-aged persons.

    Methods: A total of 40,803 subjects aged 40–59 years living in the community who were free of prior diagnoses of cardiovascular disease and cancer and who completed a food frequency questionnaire were followed from 1990–1992 to the end of 2001 in the Japan Public Health Center-based Prospective Study.

    Results: After 468,472 person-years of follow-up, 251 coronary heart disease incidents were documented. Coronary heart disease and definite myocardial infarction were inversely associated with dietary intake of folate, VB6 and VB12 after adjustment for age and sex, but the associations were attenuated after further adjustment for smoking, dietary and other cardiovascular risk factors. However, among non-multivitamin supplement users, multivariable hazard ratios (95% confidence intervals) in the highest vs. lowest quintiles of VB6 intake were 0.60 (0.37–0.97) for total coronary heart disease and 0.52 (0.29–0.91) for definite myocardial infarction, and the inverse associations with VB12 were marginally significant. The combination of below-median intake of three vitamins or of only B6 conferred a twice excess risk of total coronary heart disease.

    Conclusions: Dietary intake of VB6 was associated with a reduced risk of coronary heart disease among middle-aged non-multivitamin supplement users. Dietary folate and VB12 were also suggested to be protective factors for coronary heart disease.

    We investigated the photonic crystal structure inside iridescent scales of the weevil Lamprocyphus augustus. By combining a high-resolution structure analysis technique based on sequential focused ion beam milling/scanning electron microscopy imaging with theoretical modeling and photonic band structure calculations we discovered a natural 3-dimensional photonic structure with a diamond-based crystal lattice operating at visible wavelengths. Moreover, we found that within individual scales the diamond-based structure is assembled in form of differently-oriented single-crystalline micrometer-sized pixels with only selected lattice planes facing the scales’ top-surface. A comparison of results obtained from optical micro-reflectance measurements with photonic band structure calculations reveals that it is this sophisticated micro-assembly of the diamond-based crystal lattice that lends Lamprocyphus augustus its macroscopically near angle-independent green coloration.

    We examine the possibility to employ neutrinos to communicate within the galaxy. We discuss various issues associated with transmission and reception, and suggest that the resonant neutrino energy near 6.3 PeV may be most appropriate. In one scheme we propose to make Zo particles in an overtaking e+ - e- collider such that the resulting decay neutrinos are near the W- resonance on electrons in the laboratory. Information is encoded via time structure of the beam. In another scheme we propose to use a 30 PeV pion accelerator to create neutrino or anti-neutrino beams. The latter encodes information via the particle/anti-particle content of the beam, as well as timing. Moreover, the latter beam requires far less power, and can be accomplished with presently foreseeable technology. Such signals from an advanced civilization, should they exist, will be eminently detectable in neutrino detectors now under construction.

    Plant domestication represents an accelerated form of evolution, resulting in exaggerated changes in the tissues and organs of greatest interest to humans (for example, seeds, roots and tubers). One of the most extreme cases has been the evolution of tomato fruit. Cultivated tomato plants produce fruit as much as 1,000 times larger than those of their wild progenitors. Quantitative trait mapping studies have shown that a relatively small number of genes were involved in this dramatic transition, and these genes control two processes: cell cycle and organ number determination. The key gene in the first process has been isolated and corresponds to fw2.2, a negative regulator of cell division. However, until now, nothing was known about the molecular basis of the second process. Here, we show that the second major step in the evolution of extreme fruit size was the result of a regulatory change of a YABBY-like transcription factor (fasciated) that controls carpel number during flower and/or fruit development.

    Materials are typically ductile at higher temperatures and become brittle at lower temperatures. In contrast to the typical ductile-to-brittle transition behavior of body-centered cubic (bcc) steels, we observed an inverse temperature dependence of toughness in an ultrahigh-strength bcc steel with an ultrafine elongated ferrite grain structure that was processed by a thermomechanical treatment without the addition of a large amount of an alloying element. The enhanced toughness is attributed to a delamination that was a result of crack branching on the aligned {100} cleavage planes in the bundles of the ultrafine elongated ferrite grains strengthened by nanometer-sized carbides. In the temperature range from 60° to –60°C, the yield strength was greater, leading to the enhancement of the toughness.

    Obesity and metabolic syndrome are increasingly recognized as major risk factors for cardiovascular disease. Herein we show that Krüppel-like transcription factor 5 (KLF5) is a crucial regulator of energy metabolism. Klf5+/- mice were resistant to high fat–induced obesity, hypercholesterolemia and glucose intolerance, despite consuming more food than wild-type mice. This may in part reflect their enhanced energy expenditure. Expression of the genes involved in lipid oxidation and energy uncoupling, including those encoding carnitine-palmitoyl transferase-1b (Cpt1b) and uncoupling proteins 2 and 3 (Ucp2 and Ucp3), was upregulated in the soleus muscles of Klf5+/- mice. Under basal conditions, KLF5 modified with small ubiquitin-related modifier (SUMO) proteins was associated with transcriptionally repressive regulatory complexes containing unliganded peroxisome proliferator–activated receptor-δ (PPAR-δ) and co-repressors and thus inhibited Cpt1b, Ucp2 and Ucp3 expression. Upon agonist stimulation of PPAR-δ, KLF5 was deSUMOylated, and became associated with transcriptional activation complexes containing both the liganded PPAR-δ and CREB binding protein (CBP). This activation complex increased the expression of Cpt1b, Ucp2 and Ucp3. Thus, SUMOylation seems to be a molecular switch affecting function of KLF5 and the transcriptional regulatory programs governing lipid metabolism.

    We report here the radial compression of a large number of antiprotons (~5×105) in a strong magnetic field under ultrahigh vacuum conditions by applying a rotating electric field. Compression without any resonant structures was demonstrated for a range of frequencies from the sideband frequency of 200 kHz to more than 1000 kHz.     This paper describes the crystal structure, magnetic properties, and photoreversible magnetic properties of CoII3[WV(CN)8]2(pyrimidine)4·6H2O. We found that complexes of this formula had two types of crystal structures ([{CoII(pyrimidine)2}2{CoII(H2O)2}{WV(CN)8}2]·4H2O (1) and [{CoII(pyrimidine)(H2O)}2{CoII(H2O)2}{WV(CN)8}2](pyrimidine)2·2H2O (2)). These two structures had similar metal-organic frameworks but differed in the coordination environment around Co1, i.e., Co1(NC)4(pyrimidine)2 in 1 and Co1(NC)4(pyrimidine)(H2O) in 2. In 1, a temperature-induced phase transition from the CoII (S = 3/2)-NC-WV (S = 1/2) [high-temperature (HT)] phase to the CoIII (S = 0)-NC-WIV (S = 0) [low-temperature (LT)] phase was observed due to a charge-transfer-induced spin transition. However, 2 did not exhibit such a phase transition. When the LT phase of 1 was irradiated by 840 nm light, ferromagnetism with a Curie temperature of 40 K and magnetic coercive field of 12 kOe were observed. UV–vis reflectance and infrared measurements suggested that the LT phase optically transited to the photoinduced (PI) phase, which had a similar valence state as the HT phase, through the metal-to-metal charge-transfer (WIV → CoIII) band. In contrast, when the back metal-to-metal charge transfer (CoII → WV) band of the PI phase was excited by 532 nm light, the reverse phase transition from the PI phase to the LT phase occurred, and the spontaneous magnetization decreased.

    Massive stars end their short lives in spectacular explosions―supernovae―that synthesize new elements and drive galaxy evolution. Historically, supernovae were discovered mainly through their 'delayed' optical light (some days after the burst of neutrinos that marks the actual event), preventing observations in the first moments following the explosion. As a result, the progenitors of some supernovae and the events leading up to their violent demise remain intensely debated. Here we report the serendipitous discovery of a supernova at the time of the explosion, marked by an extremely luminous X-ray outburst. We attribute the outburst to the 'break-out' of the supernova shock wave from the progenitor star, and show that the inferred rate of such events agrees with that of all core-collapse supernovae. We predict that future wide-field X-ray surveys will catch each year hundreds of supernovae in the act of exploding.

    WNIによりますと、5月27日(火)午後3時頃、フィリピンの東海上で台風5号ナクリー(カンボジアの花の名前)が発生したそうです。

    今後は北上して、小笠原諸島に影響すると予想されています。現在のところ、本州への影響はない見込みです。

    腹痛

    どうもお腹をこわしたようで、久しぶりに、定期的に腹が痛くなってはトイレに駆け込むというのを繰り返しております。しかも、こういう時に限って正午から二つも外せないミーティングがありまして、途中、あまりの脱線加減にかなり苛ついたのはそのせいでございます。その後、仕方がないので、早退してきました。

    これでも午前中よりは多少良くなっておりまして、只今、寝床で眠れないまま、横になっております。帰りがけに、御大と秘書さん宛で早退願をメールで投げたのですが、一向に返事が来る気配もなく、これで承認されなかったら無断早退にでもなるのかと、妙な期待と不安が入り交じっております。

    ということで、探査機が無事に火星に着陸したり、某アナウンサーが練炭自殺したりしておりますが(関係ないか…)、エントリーはお休み…。何卒、ご了承下さいませ。

    gooブログに移って、1,000日目となりました。ありがとうございます。

    …というかですね、せっかく、日付指定までしてエントリー準備していたのに、結局当日にはエントリー出来ず仕舞いでした。
    思えば、職場のサーバーで自分のページの更新情報を流し始めて以来、多少の中断をはさみながらも続けてきまして、途中でgooブログに移行して、その移行から1,000日ということですので、実際の所は2000年の年末からなのでかれこれ…すみません、中断をはさむと訳が分かりません
    てことで、実際の所、gooブログに移行して1,001日となってしまいましたが、一応、1,000日を超えたということで…

    これからも独り言や愚痴や戯言や噂ばっかりですが、今後とも宜しくお願いします。

    火事

    只今、自宅です。先程、着いてやっと落ち着いたところです。で、やたら消防車がたくさん家の前の通りを通過すると思ったら、通過じゃなくて集まってまして、えらく近所でかなり大きな火事です。

    そりゃもうかなり大火事です。そこらじゅう消防車だらけで、えらく臭いと思ったら、火事じゃなくて消防車の排気ガスで煙ってました。おまけに、どこからわいてくるのか、すごい数の野次馬です。

    なんか、当分の間、騒々しそうです。いやぁ、騒ぎの前に帰宅出来て助かりました。
