Chemotherapy resistance and overexpression of proteins 14-3-3 - Led Tube Light Bulb

Certain proteins, such as 14-3-3, conserve their basic functions ofcell cycle control in diverse organisms, from worms to humans. In astudy led by Julian Cer0n and Simo Schwartz Jr, researchers fromthe Bellvitge Biomedical Research Institute (IDIBELL) and theResearch Institute of Vall d'Hebron (VHIR) respectively, havedescribed germ line functions of par-5, which is one of the two14-3-3 proteins existing in Caenorhabditis elegans , worms used as experimental model in genetic studies. Theoverexpression of the 14-3-3 proteins is related to the resistanceof tumors to chemotherapy, which could have implications forclinical practice. Researchers found that par-5 gene, as its human homologs, isrequired for DNA damage response in C. elegans validating the model to investigate chemotherapies and geneticmodifications since 14-3-3 proteins are therapeutic targets in cancer. The results of this research have been published in the latestissue of Journal of Cell Science. The powerful genetic tools of C. elegans have allowed a precise functional dissection of the single 14-3-3protein present in their germline. The researchers have discoveredthat par-5 is not only necessary for proper cell cycle regulation, but also to prevent the accumulation of endogenous DNA damage andgenomic instability. Moreover, this study reveals that par-5 is required for DNA repairresponse when it is damaged by chemicals or ionizing radiation. Insuch response, the researchers propose a model where PAR-5regulates CDK-1 phosphorylation to stop the cell cycle and repairthe damage induced by chemotherapeutic agents. The overexpression of the 14-3-3 protein has been related tochemotherapy resistance in cancer cell lines while itsdownregulation sensitizes cells to therapy-induced cell death. Therefore, this study in C. elegans provides the basis for a model to study chemotherapy response inthe context of a whole living organism. Regulators proteins 14-3-3, evolutionarily conserved, bind tosignaling proteins and affect their stability, activity or cellularlocalization. So, they are involved in the regulation of variouscellular processes, including apoptosis, the cell cycle and stress response. In addition, the researchers found that par-5 is required for cellcycle arrest in response to replicative stress and ionizingradiation. Additional References Citations.

Evidence of hidden heart disease in hypertensive african-americansrevealed by study

A Wayne State University School of Medicine study has found that anoverwhelming majority of African-American patients with hypertension also suffered hidden heart disease caused by high blood pressure even though they displayed no symptoms. The study - "Subclinical Hypertensive Heart Disease inAfrican-American Patients with Elevated Blood Pressure in anInner-City Emergency Department" - was conducted by Phillip Levy,M.D., M.P.H., associate professor of Emergency Medicine, and wasrecently published online in Annals of Emergency Medicine. Nine of every 10 patients tested suffered hidden heart damagecaused by high blood pressure, the study found. While slightly morethan 93 percent of 161 patients in the study had a history ofhypertension, 90.7 percent tested positive for hidden hypertensiveheart disease.

None of them knew their high blood pressure wasaffecting their hearts and did not show any symptomatic signs ofheart disease. "These results present a tremendous opportunity to screen for heartdisease before it becomes symptomatic, especially in a populationwith high rates of hypertension," Levy said. "If we can detectincipient heart disease early, we have a better shot at treating itbefore it turns into a full-blown health emergency. Our study isalso a strong reminder that emergency patients with chronic disease- in this case, hypertension - are generally a high-risk group." The patients were enrolled in the study after appearing at theemergency room of Detroit Receiving Hospital. They did not come tothe hospital for heart disease symptoms.

Once enrolled in thestudy, they underwent echocardiograms, which revealed thehypertensive heart disease. Of the total 161 patients, 93.8 percentwere inner-city African-Americans; 51.6 percent were male. The meanage of the patients enrolled was 49.8 years. Most of the patients (93.8 percent) had a history of high bloodpressure and were aware that they had the condition, but only 68.3percent were receiving treatment. Led Growing Lights

Of those found to have hidden heart disease, the majority werediagnosed with diastolic dysfunction, defined as the heart'sinability to adequately pump blood. Levy said the echocardiogramsfound the presence of subclinical hypertensive heart disease"ubiquitous." He noted that hypertension is commonplace in the United States, andaffects more than 76 million adults. The prevalence of thecondition is higher in African-Americans, who are at "tremendousrisk" for pressure-related consequences of hypertension, especiallythe premature onset of damage to and impairment of heart function. According to the U.S. Dimmable Led Aquarium Light

Centers for Disease Control and Prevention,228.3 in every 100,000 Michigan residents 35 and older died ofhypertension-related causes in 2009. In African-Americans, the ratewas 381.9 deaths for every 100,000, and in whites it was 211. AllMichigan rates were higher than national statistics. In rates ofhypertension hospitalizations of Michigan residents 65 and olderwho are Medicare beneficiaries, African-Americans had higher rates (14hospitalizations per 1,000 Medicare beneficiaries) than whites (3.6per 1,000). Again, both rates were higher than national numbers. China Led Aquarium Lighting

Since subclinical heart disease is unlikely to be detected in suchhypertensive patients until the damage manifests in visiblyrecognized symptoms, the early identification of the condition "hasemerged as an important aspect of secondary cardiovascular diseaseprevention," Levy said. Emergency room physicians may underestimate the prevalence ofhidden hypertensive heart disease in inner-city African-Americans,who are considered an especially high-risk group and who rely onemergency rooms for treatment because of lack of access to primarycare physicians. "Emergency physicians are uniquely positioned to lessen the overallimpact of chronic high blood pressure in at-risk communities," Levysaid. "Blood pressure readings are taken for every patient in theER. By not just taking in new information but also acting on it, wecan substantively contribute to much-needed secondary diseaseprevention efforts." In 2010, the CDC reported the financial burden of hypertension inthe U.S.

was $76.6 billion in health care, medication and misseddays of work. Recognizing the likelihood of previously unrecognized subclinicalhypertensive heart disease prevalence in African-Americans holdstherapeutic promise that could reduce the adverse outcomes, Levysaid. Blacks progress from hidden to symptomatic stages ofleft-ventricular dysfunction more rapidly than other populationgroups, and the mean age of blacks admitted to hospitals with heart failure is much lower than that of whites (63.6 years versus 75.2 years). "While we must recognize the risk that exists for these patients,we should not expect emergency departments to perform the furtherstudies needed to identify subclinical end-organ damage," Levysaid. "Emergency departments should focus on identification ofpoorly controlled hypertension - whether or not it was the primaryreason for the emergency visit - and hospital systems, especiallythose where high disease prevalence exists, should have somecoordinated mechanism where patients can be referred for follow-up.Moreover, at that follow-up, a mechanism should exist to performeffective intervention, including risk stratification, even ifpatients lack insurance."
